INTRODUCTION

RAPITESTTM Malaria Pf/Pan is a rapid, quantitative immunoassay for the detection of P. falciparum specific histidine rich protein – 2 (Pf HRP-2) and Pan malaria specific Parasite lactate dehydrogenase (pLDH) in human blood as an aid in the diagnosis of malaria infection. This test is also used to differentiate the P. falcipuram and the other malarial species. This test is very sensitive and rapid. Results are interpreted in less than 20 minutes. Test results are read visually without any instruments.

 PRINCIPLE:

RAPITESTTM malaria Pf/Pan is a membrane based two site immunochromatographic test, which is coated with monoclonal antibody specific to HRP-2 of P.falcipuram on the test line 1 and monoclonal antibody specific to pLDH of Pan malarial species (P.Vivax, P. malariae and P. ovale) on the test line 2. The control region “C” is coated with goat anti mouse IgG. The test sample moves further on the membrane after addition of running buffer. The gold conjugated monoclonal anti Pf HRP-2 and Monoclonal anti pan-pLDH antibodies complexes with the proteins in the sample. This complex moves further on the membrane to the test region, leading to the formation of pink/purple color line on the test region which confirms the positive reaction. Both the bands will appear at the test region for P. falcipuram positive positive samles and only one band will appear at the test region for non P. falcipuram positive samples. Absence of these bands on the test regions indicates no reaction. The unreacted gold conjugate and color beads move further on the membrane at the control region and forms pink color line. The control line serves to validate the test result. No band at control region “C” indicates the test is INVALID.
